        <p>Обоснование. Благодаря своей высокой антиоксидантной активности кофейная кислота обладает рядом положительных фармакологических эффектов. Для количественного определения кофейной кислоты в различных препаратах в настоящее время используются такие физико-химические методы как жидкостная и газовая хроматографии, УФ-спектрофотометрия, капиллярный электрофорез с масс-спектрометрией. Однако, перечисленные методы являются достаточно сложными и многостадийными, требуют специализированного оборудования и значительных затрат времени. Это определяет потребность поиска новых способов определения кофейной кислоты, отличающихся простотой, чувствительностью и эффективностью ее обнаружения. В настоящей работе нами выявлен эффект излучения квантов света при окислении кофейной кислоты пероксидазой хрена, данная реакция явилась предметом нашего изучения. 
 
Цель. Исследовать некоторые основные показатели реакции люминесценции при окислении кофейной кислоты пероксидазой хрена и оценить возможность определения кофейной кислоты с помощью предложенного люминесцентного метода. 
 
Материалы и методы. В экспериментах использовали реактивы высокой степени чистоты: пероксидазу из корней хрена (horseradish root peroxidase – HRP), кофейную кислоту, раствор перекиси водорода. Водные растворы реактивов для проведения экспериметов готовили in situ в деионизированной воде. Интенсивность и динамику световой эмиссии реакций регистрировали на люминометре. Люминесцентные сигналы измеряли в режиме одно измерение в секунду и выражали уровень световой эмиссии в относительных единицах. 
 
Результаты. Установлено, что в присутствии пероксида водорода HRP катализирует окисление кофейной кислоты с величиной Кm = 1.4 × 10-4 M и обеспечивает реакцию светоизлучения в широком диапазоне рН с максимумом световой эмиссии при pH 8. В экспериментах in vitro показана применимость катализируемой HRP люминесцентной реакции для быстрого выявления и определения концентрации кофейной кислоты в водной среде. Установлено, что линейное увеличение регистрируемого светового сигнала наблюдается в диапазоне концентраций аналита 20–200 мкМ. 
 
Заключение. Выявленная реакция светоизлучения является перспективной аналитической платформой и открывает возможности создания на ее основе новых люминесцентных сенсоров для тестирования кофейной кислоты в различных препаратах и экстрактах из биологических источников.</p>

      <trans-abstract xml:lang="en">
        <p>Background. Due to its high antioxidant activity, caffeic acid has a number of positive pharmacological effects. For the quantitative determination of caffeic acid in various preparations, some physicochemical methods, such as liquid and gas chromatography, UV spectrophotometry, capillary electrophoresis with mass spectrometry, are currently used. However, it is worth noting, that these methods are quite complex and multi-stage, require specialized equipment and significant time costs. This determines the need to search for new methods for determining caffeic acid, distinguished by simplicity, sensitivity and efficiency of its detection. In this work, we have revealed the effect of light quantum emission during the oxidation of caffeic acid by horseradish peroxidase; this reaction was the subject of our study. 
 
Purpose. To investigate some of the main performances of the luminescence reaction during the oxidation of caffeic acid by horseradish peroxidase and to evaluate the possibility of determining caffeic acid using the proposed luminescence method. 
 
Materials and methods. The experiments were performed using high-purity reagents: horseradish root peroxidase (HRP), caffeic acid, and hydrogen peroxide solution. Aqueous solutions of the reagents for the experiments were prepared in situ in deionized water. The intensity and dynamics of the light emission of the reactions were recorded using a luminometer. Luminescent signals were measured in the mode of one measurement per second and the level of light emission was expressed in relative units. 
 
Results. It was found that HRP catalyzes the oxidation of caffeic acid in the presence of hydrogen peroxide with a Km value of 1.4 × 10-4 M and provides a light emission reaction in a wide pH range with a maximum light emission at pH 8. In vitro experiments demonstrated the applicability of the HRP-catalyzed luminescent reaction for rapid detecting and determining the concentration of caffeic acid in an aqueous medium. A linear increase in the recorded light signal is ensured in the analyte concentration range of 20–200 μM. 
 
Conclusion. The obtained results indicate that the discovered light emission reaction is a promising analytical platform and opens up the possibilities of creating new luminescent sensors on its basis for testing caffeic acid in various preparations and extracts from biological sources.</p>
